The REPAMO network (network ofmollusc pathology), created in 1986, ensures the survey of shellfish health status along French coasts according? to European Directives 91/67/EEC and 95/70/EEC. The aims of the network are 1 - to prevent the introduction and spread of exotic pathogens, 2-to study the means to decrease the impact of pathogens already present and to survey their evolution. In 2003, the REPAMO network focused on the survey oflisted diseases (bonamiosis and marteiliosis) in two areas under agreement process (Banc de Granville and zone X) and on the study of abnormal morlalities. Number of reported abnormal morlalities has increased in 2003. This rise can be partly explained by the recent network restructuring, but also by the summer heatwave and...

L'amélioration des performances de l'huître plate européenne Ostrea edulis par la sélection génétique ArchiMer

The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae.

Bedier, Edouard; Claude, Serge; Simonne, Charlotte; D'Amico, Florence; Palvadeau, Hubert; Guilpain, Patrice; Le Gall, P; Pien, S.

Bedier, Edouard; Fleury, Pierre-gildas; Langlade, Aime; Haure, Joel; Le Roux, A.; Ropert, Michel; Rauflet, Fabienne; Soletchnik, Patrick.
The "sedimentary risk" has been identified since the mid-nineties in the Marennes Oléron Bay. The proximity of the sediment (from 0 to 15cms) increases the mortality rate. Depending on the year, mortality can be 10 to 20 % higher than in "high" conditions (50 to 70cms). These zootechnic results, gathered since 1996 on the working site of Ronces – Perquis, lead to the research of a pathogenic agent or a toxic substance released by the sediment. Studies conducted between 2002 and 2003 [12, 18] did not allow the identification of any pathogenic agents and ruled out the infectious risk. In 2004, the chemical risk (sulphides, ammoniacal nitrogen) was studied on 4 sites (3 working sites + Quiberon). In 2004, the "on-bottom/off bottom" mortality model of the...

The phenomenon of summer mortalities of the Pacific cupped oyster Crassostrea gigas is observed in France since the beginning of the 90's. These mortalities are classically associated to younger stages and summer temperatures and occur during the reproduction period of the animal. The studies undertaken during the project Morest (2001-2005), showed that a number of biological and environmental risks, the interaction of them induces lethal stresses for animals in physiological weakness, exists in summer. Among these risks, that which is withstood by the oysters bred close to the sediment proved to be significant in intertidal areas. In Quiberon bay (southern Brittany, France), the breeding practices of sowing oysters in deep water is thus likely to expose...

Validation in commercial conditions of the response to selection of the European flat oyster Ostrea edulis for resistance to Bonamia ostreae ArchiMer
Bedier, Edouard; Langlade, Aime; Angeri, Stéphane; Brizard, Raphael; Nerlovic, V.; Glize, Philippe; Haffray, Philippe.
Since the outbreaks of the parasitosis with Marteilia refringens and Bonamia ostreae in the last 60's, the French production of the European flat oyster dropped from 20000 mT to less than 1500 mT currently, leaving the French shellfish farming in situation of virtually monoculture of the Pacific cupped oyster Crassostrea gigas. To sustain this traditional culture, Ifremer has developed, at the experimental level, a genetic selection for a better tolerance to Bonamia ostreae which led to an improvement of survivals. The transfer of these results to the business scale requires the control of the reproduction in hatchery and the adaptation of the cycle of production to the constraints of seedlings in deep water as practised in Brittany (France). That's why a...

Microsatellite-associated heterosis in hatchery-propagated stocks of the shrimp Penaeus stylirostris ArchiMer
Bierne, Nicolas; Bezuart, Ivan; Vonau, Vincent; Bonhomme, François; Bedier, Edouard.
Correlation between DNA microsatellite heterozygosity and growth rate was investigated in two hatchery-propagated stocks of the shrimp Penaeus stylirostris, which had been genetically isolated from wild founders for 17 generations, and from each other for five more generations. presumed demographic history of these populations suggested that they were maintained at small effective population sizes. and this was confirmed by significant changes in allelic frequencies between the two stocks. Despite a small sample size (n = 60) and a small number of loci screened, a significant positive correlation was detected between microsatellite tri-locus heterozygosity and growth rate in one of the two stocks (r = 0.3, P = 0.02). In the other stock (n = 48), the...

Response to divergent selection for resistance to summer mortality in Pacific oyster spat : genetic parameters and correlated responses with subsequent survival and growth ArchiMer
Boudry, Pierre; Bedier, Edouard; Ernande, Bruno; Degremont, Lionel.
The multidisciplinary program "MOREST" aims to improve our understanding of causes of summer mortality in Crassostrea gigas juveniles in France and to reduce its impact on oyster production. As part of this program, four successive generations of families were bred between 2001 and 2005 in our research hatchery in La Tremblade. Survival and growth were recorded in the field in three sites along the French coast (Ronce, Rivière d'Auray and Baie des Veys). The first generation (44 full-sib families nested within 17 half-sib families) was tested during summer 2001 to assess to what extent genetic variability exists for spat survival. An important genetic basis was found (estimated narrow-sense heritability among sites = 0.83 ± 0.40). In contrast, lower...

Genetic variation and trade-offs for reproduction ans survival in the Pacific oyster Crassostrea gigas ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Pouvreau, Stephane; Normand, Julien; Ernande, Bruno.
To date, the most significant method to genetically reduce reproductive effort in the Pacific oyster (Crassostrea gigas) has been through the production of triploids, especially since the development of tetraploid lines allowing the breeding of 'natural' triploids. Gametogenesis of triploid oysters is strongly reduced compared with diploids, although they are not fully sterile and can produce viable gametes and some progenies when crossed with diploids. Reduced reproductive allocation and higher heterozygocity are commonly proposed as the main reasons why triploids often present superior yield compared with diploids. Temperature and food availability are known to favour gametogenesis in triploids, but the existence of genetic variation for this trait...

Selective breeding to improve resistance against summer mortality in the Pacific oyster Crassostrea gigas : results after 3 generations ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Samain, Jean-francois.
Summer mortality of adults and juveniles has been reported in the Pacific oyster, Crassostrea gigas, for many years and in several countries. The French multidisciplinary program "Morest" aims to investigate the causes of the summer mortality in Crassostrea gigas. Within this program, we designed multi-site field experiments to assess to what extent genetic variability exists for summer mortality in French populations of C. gigas and to determine whether selective breeding could improve survival.

Etude comparative en cytométrie en flux des fonctions estérases et du métabolisme oxydatif d'hémocytes d'huîtres saines, parasitées, sensibles et résistantes au protozoaire Bonamia ostrea ArchiMer
Cochennec, Nathalie; Garcia, Sandrine; Bedier, Edouard.
In order to fight Bonamiosis, the applicable means are very limited and mostly based on a quick and precocious diagnostic aimed at avoiding the propagation of the disease and a rise in the "resistance" through genetic selection. Various works are conducted in order to understand the oysters' defence mechanisms against this parasite, and then to develop new prophylaxis and fighting methods. In our laboratory, we focus on cellular compartments, the circulating haemocytes, in which B. ostreae multiplies, and their interaction with the parasite.

Summer mortality of hatchery-produced Pacific oyster spat (Crassostrea gigas). II. Response to selection for survival and its influence on growth and yield ArchiMer
Degremont, Lionel; Bedier, Edouard; Boudry, Pierre.
Response to divergent selection for "high" and "low" survival during the summer period, from July to October, was investigated in juvenile (six-month-old) Pacific oysters, Crassostrea gigas, by producing two sets of progenies in 2002 (Generation 2) and three sets of progenies in 2003 (Generation 3). A strict between-family approach was used and resistance of these selected progenies to summer mortality was assessed in three sites along French coasts, to determine their response to selection and estimate realized heritability of the trait. A significant difference in survival was observed between the "high" and "low" selected groups at all sites for all sets of progenies, indicating a significant genetic component. High realized heritabilities for survival...

Degremont, Lionel; Bedier, Edouard; Soletchnik, Patrick; Ropert, Michel; Huvet, Arnaud; Moal, Jeanne; Samain, Jean-francois; Boudry, Pierre.
Summer mortality has been reported in the Pacific oyster, Crassostrea gigas, for many years in different parts of the world. The causes of this phenomenon are complex. The multidisciplinary program "MOREST", coordinated by IFREMER, was initiated to understand the causes of summer mortality of Crassostrea gigas juveniles in France and to reduce its impact on oyster production. Within this program, three successive groups of bi-parental families were bred in a hatchery in 2001 and placed in the field during summer in three sites (Ronce, Riviere d'Auray, and Baie des Veys). This paper reports the relative importance of family, site and field placement timing for three characters of major importance for oyster production: survival, growth, and yield. At the...

The French project "Morest", coordinated by IFREMER, aims to a better understanding of the causes of summer mortalities in juvenile oysters by a multidisciplinary approach. Within this program, a first generation (G1) was bred in 2001 to assess to what extent genetic variability exists for this trait in French populations. 17 half-sib families (HSF) including 44 full-sib families (FSF) were obtained in the G1, and each FSF was reared in three sites along the French coast during the summer period. Significant differences in survival were observed among HSF, clearly indicating a genetic basis for survival. Estimation of heritability for survival was very high: 0.81  0.29. In 2002, 3 "good" HSF and 3 "bad" HSF were selected on their survival...

The French project "Morest", coordinated by IFREMER, aims to understand the causes of the summer mortalities in Crassostrea gigas. In 2001, three sets of families were bred following a nested half-sib mating design. 17 half-sib families (HSF) were obtained in this first generation (G1) and reared in 3 sites. Significant differences in survival were observed among HSF, and some HSF showed high levels of mortality in all sites, clearly indicating a genetic basis for survival. In 2002, a second generation (G2), including divergent selection and inbred lines, was constituted. Monitoring of survival and growth of G2s were the same as in 2001. In the scope of the Morest program, 3 series of bi-parental families of cupped oyster consisting of full and half-sib have been created in 2001, and then studied on a regular basis on several sites over the summer period. In October, major survival differences had been demonstrated between the 3 families. The average survival rate for the half-sib families monitored on the three sites varies from 17 to 97 %. Some of them showed high mortality for the three sites, indicating a strong genetic basis (h2 = 0.81 - 0.29) to the variations observed regarding survival. To a lesser extent, growth differences between the studied half-sib families indicate a weaker heritability for this character. ....

Effects of age and environment on survival of summer mortality by two selected groups of the Pacific oyster Crassostrea gigas ArchiMer
Degremont, Lionel; Boudry, Pierre; Ropert, Michel; Samain, Jean-francois; Bedier, Edouard; Soletchnik, Patrick.
This study investigated the effects of age (first or second summer of life) and environment (three study sites on French coasts) on summer mortality in Crassostrea gigas, using three generations of oysters produced during the Morest project. For each generation, two groups of oysters, selected either as 'resistant' (R) or 'susceptible' (S) to summer mortality at the age of 6 months in Rivière d'Auray (RA, South Brittany), were evaluated. In Ronce (Marennes-Oléron Bay) and RA, R oysters had low mortality during their first and second summers, while mortality was higher during the second summer in Baie des Veys (BDV, Normandy). In RA, S oysters exhibited higher mortality during their first summer than during their second, while the opposite result was found...
